Th1/Th2细胞因子mRNA的表达与角膜缘移植排斥的关系

【摘要】 目的研究细胞因子(IL-2、IFN-r、IL-4、IL-10)在大鼠角膜缘移植模型中的动态表达,探讨其与移植排斥反应发生的关系。方法将大鼠随机分为同种异体移植组(BN→Lewis)、同体移植组(Lewis→Lewis),并以健康大鼠作为正常对照,用实时定量PCR方法,在术后1、3、7、14、30d分别检测角膜缘组织中细胞因子(IL-2、IFN-r、IL-4、IL-10) mRNA的表达,并观察大鼠眼表变化,判断排斥反应的发生。结果IL-2及IFN-r在异体移植组的表达较同体移植组明显升高,除术后第1d外,各时间点差异均有统计学意义(P<0.01)。术后7d两组同时达高峰,随后逐渐下降。IL-4及IL-10 mRNA在两组表达量较低,各时间点差异无统计学意义(P>0.05)。结论在大鼠异体角膜缘移植排斥过程中,Th1细胞因子IL-2、IFN-γ发挥了重要的作用。

【Abstract】 ObjectiveThe reject reaction following limbal transplantation is a main cause of surgery failure,however,the immunoregulation mechanism of reject remains unclear now.A dynamic change of expression of cytokine (IL-2,IFN-r,IL-4,IL-10) in the corneal limbus was investigated after limbal transplantation in this study to explain their relationship with rejection.MethodsSixty-four Lewis rats were assigned to allograft group(BN→Lewis) and isograft group(Lewis→Lewis).The right eyes of all receipter rats received limbal transplantation,and 4 healthy rats(8 eyes)were as blank controls.The expressions of IL-2,IFN-r,IL-4,IL-10mRNA in limbus were detected by real-time RT-PCR at postoperative day( POD ) 1,3,7,14 and 30,and the manifestation of rejection was observed on clinical scoring.Results83.3% allografts appeared clinical rejection at 5-10 days (median 7 days) after transplantation,whereas none of isografts ever did(χ2=17.143,P=0.000).Severe edema,opacity,vascularization of cornea and dense infiltration of inflammatory cells in the grafts were seen in allograft group.Mild congestion,vascularization and few inflammation cells in isografts were seen in isografts group.The expression of IL-2 and IFN-rmRNA in allograft group was significantly enhanced compared with that in isograft group.There was a significant difference between two groups (P<0.01) apart from POD1.The factors above mentioned all reached the peak on POD 7 (IL-2/β-actin: 0.120 1±0.005 2,IFN-r/β-actin: 0.158 1±0.009 3)and then gradually descended.The expression of IL-4 and IL-10mRNA was not different between two groups at any time point(P>0.05).ConclusionCytokine of Th1 (IL-2 and IFN-r) may play important roles during limbal allograft rejection in rats.
