坐骨神经慢性挤压伤大鼠背根神经节GAT-1的表达

【摘要】 目的观察背根神经节(DRG)γ-氨基丁酸转运体-1(GAT-1)在大鼠坐骨神经慢性挤压伤(CCI)后的表达,探讨慢性神经痛的发生机制。方法60只雄性SD大鼠随机分成空白对照组(Naive组)、假手术组(Sham组)和坐骨神经结扎组(CCI组,又分为结扎后3d组、7d组、14d组和28d组)。分别在各自的时间点灌注取材L4~5背根神经节,用免疫组织化学方法观察GAT-1表达。结果CCI组(3、7和14d)背根神经节GAT-1的免疫阳性神经元数和染色深度均比假手术组和空白对照组显著增加(P<0.05和P<0.01)。结论坐骨神经慢性挤压伤后背根神经节GAT-1表达增加,这可能与神经性疼痛的病理过程相关。

【Abstract】 Objective To investigate the expression of γ-aminobutyric acid transporters-1(GAT-1) in the spinal cord and dorsal root ganglion(DRG) neurons in rats with chronic sciatic nerve injury(CCI).Methods Immunocytochemistry was employed to investigate the GAT-1 expression in L_ 4-5 DRG neurons in CCI rats on the 3rd,7th,14th,and 28th day of nerve injury,which was compared with that in naive and sham operative rats.Results There was a significant increase in the numbers and grey level of GAT-1 immunoreactive cells in CCI 3d,7d,14d and 28d groups compared with those in naive or sham operation groups(P<0.05 or P<0.01).Conclusion The expression of GAT-1 is increased after CCI,which may play a role in the pathologic process of chronic nerve pain.
