一种新型生物-磁双重靶向纳米材料的制备
Preparation of a novel bio-magnetically targeted nano-materials
【摘要】 目的制备用于肿瘤靶向治疗的新型生物-磁双重靶向纳米材料。方法用部分还原-共沉淀法制备了四氧化三铁Fe3O4纳米材料,用硅烷偶联剂进行了表面修饰,用扫描电镜(SEM),X-射线粉晶衍射(XRD),磁强计(VSM),X射线能谱仪(EDS)等分析仪器对材料的相关性能进行了测定。结果磁纳米粒子经表面硅胶修饰后平均粒径在20nm左右,磁饱和强度在65emu/g;磁微粒子表面氨基官能团化后,表面氨基密度为0.5umol/g;磁纳米粒子表面进行组氨酸固定后,保持了组氨酸和纳米粒子的生物学及磁学特性;与肝癌单抗Hepama-1偶联后平均粒径在30nm左右,形态为椭圆球形,尚规则分布。结论新型生物-磁双重靶向纳米材料能满足载药的要求;通过化学偶联将一种优良的肝癌单抗Hepama-1固定在磁微粒子表面。
【Abstract】 [Objective] To investigate a method synthesized a new biologic-superparamagnetic nanoparticles as a new drug for targeting therapy of tumor. [Method] A functionalized superparamagnetic iron oxide nanoparticles (SPIONs) were synthesized by reduction-precipitation method, and were modified by silicane coupling agent. The nanoparticles were evaluated by diffraction (XRD), scanning electron microscope (SEM) and X-ray energy spectrometer (EDS). [Results] The detection with XRD, VSM, SEM, EDS showed that SPIONs′s mean size was about 20 nm, and did not change significantly after surface modification; The magnetic saturation was 65 emu/g; the surface amino group was 0.5 umol/g modified by amid-functional group. After fixed by histidine , SPION- Hepama-1 mean size was about 30 nm. [Conclusion] The SPIONs showed it′s a fine kind of biologic-magnetic nanoparticles for targeted drug delivery. Hepama-1, an excellent humanized monoclonal antibody directed against liver cancer, was conjugated to the SPIONs to prepare immuno-magnetic nanoparticles (IMN).
