偏头痛肝阳上亢证大鼠肾上腺蛋白质双向电泳图谱的分析

【摘要】 目的探寻偏头痛肝阳上亢证大鼠相关蛋白。方法将SD大鼠随机分成正常组及偏头痛肝阳上亢证组(简称模型组)。通过对肾上腺组织蛋白质的提取,经双向电泳,Pdquest软件分析,蛋白质点原位酶解及质谱分析。结果发现模型组与正常组大鼠肾上腺总蛋白质的分布模式非常相似,以PI3-8,分子量Mr14.4~75kD范围的蛋白质斑点分布最多。经PDQuest7.0软件分析后得出,在相同条件下识别的蛋白质斑点数分别为正常组(584±25)个,模型组(686±27)个。选取表达有显著差别的12个蛋白质斑点进行酶解和质谱分析,所得结果在网上搜寻,其中有9个点的蛋白质与之匹配。结论表明模型组与正常大鼠肾上腺蛋白质的表达具有差异,经质谱鉴定的9个蛋白质可能与偏头痛肝阳上亢证的形成有关.这为进一步研究偏头痛发病机制打下了基础。

【Abstract】 [Objective] To explore protein associated with migraine with excessive rising of liver-YANG. [Methods] SD rats were devided into the normal group and the migraine with excessive rising of liver-YANG group (the mode group) randomly. Adrenal glands proteins were extracted and then further analysed by two-dimensional gel electrophoresis, PDQuest 7.0 software analysis, In2gel protein digestion and mass spectrogram analysis. [Result] Total protein distribution patterns of the model group and the normal group were very similar. Protein spots were mainly displayed at pI3~8, Mr14.4~75 kD range, through PDQuest 7.0 software analyzing, and there were (686±27) protein spots in the mode group while (584±25) spots in the normal group. Take 11 protein spots that were notable expressed to digest and MALDI-TOF-MS analyze.Compared with the results found on the internet, 8 protein spots were matched. [Conclusion] There are differences in the model group and the normal Adrenal glands. These 8 protein spots may be concerned with the formation of migraine with excessive rising of liver-yang. This result will provide foundation for identifying the pathogenesy of migraine.
