人表皮生长因子在转基因番茄中成功表达

【摘要】 目的以转基因番茄生产人表皮生长因子(hEGF)。方法构建hEGF基因的表达载体pCAM-35S-hEGF,通过电激法转化到农杆菌菌株C58中,采用叶盘法转化番茄品系cg2002-14。结果子叶经发芽及生根诱导培养后得到了再生植株。PCR分析表明,hEGF基因已整合到番茄的基因组中。放射免疫法(RIA)测得hEGF在果实中平均含量为3.78ng·g-1。结论成功构建了hEGF基因的表达载体,获得了表达hEGF的转基因番茄植株。

【Abstract】 Aim To produce human epidermal growth factor(hEGF)in transgenic tomatos. Methods Plasmids pCAM-35S-hEGF carrying the hEGF gene were constructed and transformed via eletro-perforation to agrobacterium C58, and then transfected to tomatoes cg2001-14 via leaf disk transformation.Results Regenerated plantlets were obtained from the cotyledons after shoot and root inducing cultivations. The PCR analysis showed that the hEGF gene has been integrated into the tomato genome. Radioimmunoassay (RIA) showed that the mean yield of hEGF in the fresh fruits of the transgenic tomatoes amounted to 3.78 ng·g-1 fruit.Conclusion The hEGF gene expression vector was successfully constructed and expressed in the transgenic tomato plants.
