乳源免疫调节肽具有促进体内外淋巴细胞转化的作用

【摘要】 目的研究乳源免疫调节肽(PGPIPN)对机体外周淋巴细胞转化的影响及其机制。方法试验分体内和体外两系列实验。体内实验,取40日龄SD大鼠42只和60只18~22g昆明小鼠作为实验动物(大鼠和小鼠均♀♂各半)。用生理盐水预饲1wk后,大鼠和小鼠均随机分为3组(大鼠每组14只,小鼠每组20只,每组♀♂相等并分笼饲养)即对照组、PGPIPN2·5×10-3g·L-1剂量组和PGPIPN2·5×10-2g·L-1剂量组,它们被分别灌喂生理盐水、2·5×10-3和2·5×10-2g·L-1乳源免疫调节肽,隔天灌喂1次,大鼠每次3ml,持续30d,小鼠每次1ml,持续14d。饲养结束后,检测对照组和两剂量组间淋巴细胞转化的差别。体外实验,用上述对照组动物血液,将酪蛋白1h、2h酶解液、2·5×10-3、2·5×10-2g·L-1乳源免疫调节肽分别加入淋巴细胞培养液,以酪蛋白和生理盐水为对照,测定乳源免疫调节肽对淋巴细胞转化的直接影响。结果体内实验,饲喂乳源免疫调节肽能明显提高淋巴细胞转化的刺激指数,其中大鼠的PG-PIPN2·5×10-2g·L-1剂量组以及小鼠的PGPIPN2·5×10-3、2·5×10-2g·L-1剂量组均高于各自的对照组(P<0·05)。体外实验,不论大鼠和小鼠,酪蛋白、酪蛋白1h酶解液、酪蛋白2h酶解液、2·5×10-3g·L-1、2·5×10-2g·L-1乳源免疫调节肽均能提高淋巴细胞转化的刺激指数,幅度按上述排列逐渐增大,其中2·5×10-2g·L-1乳源免疫调节肽组高于各自的对照组(P<0·05),大鼠的2·5×10-2g·L-1免疫调节肽组也高于酪蛋白组(P<0·05),但酪蛋白组与对照组相比,差异较小。体内外实验均存在剂量依赖关系。结论免疫调节肽在体内外均能促进外周淋巴细胞的转化,而且有剂量依赖关系。

【Abstract】 Aim To investigate the effect and mechanism of immunomodulating peptide (PGPIPN) derived from β-casein on lymphocyte transformation (LT). Methods There were two series of experiments both in vivo and in vitro. In vivo, both 42 SD rats and 60 kunming mice were randomly divided into the control, PGPIPN ( 2.5×10~ -3 g·L~ -1 ) and PGPIPN (2.5×10~ -2 g·L~ -1 ) group (14 rats in each group, 20 mice in each group), which were respectively administrated with saline and the immunomodulating peptide at 2.5×10~ -3 and 2.5×10~ -2 g·L~ -1 via gastric cannula for 30 d in rats and 14 d in mice. LT in both rats and mice were respectively measured at end in vitro, culture solutions including lymphocytes, which came from animal in control group (rats: n=14, mice: n=20), were respectively added with casein 1 h hydrolysate,casein 2 h hydrolysate, PGPIPN at 2.5×10~ -3 and 2.5×10~ -2 g·L~ -1 , while saline and casein were taken as contrast. The direct effects of immunomodulating peptide on LT in both rats and mice were respectively measured. Results In vivo, LTs in both rats and mice were higher in PGPIPN groups than those in the control group, thereinto, LTs in PGPIPN ( 2.5×10~ -2 g·L~ -1 ) group of rats and in PGPIPN (2.5×10~ -3 and 2.5×10~ -2 g·L~ -1 ) groups of mice increased significantly in contrast with their control group (P<0.05).There was a dose-dependent manner between immunomodulating peptide and LT level in vitro, LT were all improved with casein, casein 1 h hydrolysate, casein 2 h hydrolysate, PGPIPN at 2.5×10~ -3 and 2.5×10~ -2 g·L~ -1 , LT levels gradually increased according to this order. In rats, LT level in the group with immunomodulating peptide at 2.5×10~ -2 g·L~ -1 was markedly higher than that in the control and casein groups (P<0.05). Compared with the control group, LT in the group with immunomodulating peptide at 2.5×10~ -2 g·L~ -1 in mice was significantly improved (P<0.05). The difference in LT level between the control group and casein group were very small in both rats and mice. Conclusion Immunomodulating peptide can promote LT both in vivo and in vitro. Moreover, there is also a dose-dependent manner between immunomodulating peptide and LT level.
