【摘要】 以龙眼“石硖”、“大乌圆”、“大广眼”、“龙荔”叶片为试材,研究了一种适宜AFLP分析的龙眼基因组DNA提取方法,其改良策略是:在细胞核被裂解前先除去多酚类等次生物质,防止多酚类的氧化,细胞裂解后再加入10%的CTAB,进一步除去杂质。此方法不仅适用于嫩叶采样,也适用于老叶采样,所提取的基因组DNA可成功进行AFLP标记分析,为龙眼的分子生物学研究提供了科学依据。更多还原

【Abstract】 A method for genomic DNA preparation of longan that is suitable to AFLP analysis has been described.The modification is to remove most of the cytoplasm components before lyses of the nuclei.After lyses of the nuclei,1/10 volume of 10% CTAB was added to remove more secondary metabolites.With the modified method,not only young leaves but also old leaves could be used to DNA preparation.The genomic DNA samples prepared with this method could be used as templates for amplified fragments length polymorphism(AFLP).更多还原
