绵羊肺炎支原体Y-98株外膜蛋白成分分析及其免疫原性研究
The SDS-PAGE analysis of immunogenicity on membrane protein of Mycoplasma ovipneumonia Y-98
【摘要】 采用TritonX-100裂解和0.6%K I提取的方法制备绵羊肺炎支原体Y-98株外膜蛋白,经12%SDS-PAGE分析,表明其分子量范围在14.4~125.9 KD之间;分别制备抗Y-98株全细胞抗原和外膜蛋白高免血清,经W estern-b lotting分析,表明外膜蛋白为绵羊肺炎支原体的主要保护性抗原,而在外膜蛋白,67.6、39.3和28.8 KD多肽为其主要保护性抗原。利用间接ELISA方法和MTT法对外膜蛋白引起的体液免疫和细胞免疫功能进行了测定,结果表明膜蛋白能够诱导机体产生高效价的抗体,免疫后7、14和28d效价分别达到了1∶24.25、1∶27.75和1∶28.75;膜蛋白具有非特异性和特异性致有丝分裂作用,在免疫前0 d及免疫后28 d对T淋巴细胞刺激的转化率分别为8.46%和18.3%。
【Abstract】 The membrane protein of Mycoplasma ovipneumonia Y-98 was extracted through TritonX-100 and 0.6 %KI lysis.The molecular weight of the protein was identified to range from 14.4 to 125.9 KD through 12 % SDS-PAGE.The high titer sera against Y-98 full cell antigen and the membrane protein were made respectively.The results of Western-blotting showed the outer membrane protein was the major protective antigen of MoP Y-98,and the 67.6,39.3,28.8 KD polypeptides were the major protective antigens of the membrane protein.The assessment of humoral and cell immune response caused by MoP membrane protein were made based on Indirect EISA and MTT,and the results showed the membrane protein could induce the secretion of high titer antibody,which were 1∶24.25,1∶27.75 and 1∶28.75at the 7th,14th and 28th day,respectively,after immunization;also the membrane protein could function as the mitogen unspecifically and specifically,which could induced the transformation of T lymphocyte and the rate of transformation were 8.46 % and 18.3 % before immunization and 28th day after immunization,respectively.
