唾液Histatin5突变体在巴斯德毕赤酵母中的优化表达

【摘要】 目的通过优化重组毕赤酵母GS115-pPICZα-A-HRP5′表达条件,提高表达水平,增强产物抗菌活性。方法将培养基甘油含量、pH、诱导前培养时间、甲醇诱导量、诱导后培养时间设置不同梯度,采用琼脂孔穴扩散法比较各梯度下表达产物抗菌活性。结果以pH6.0、甘油含量2.0%YEPG培养基、培养30h后用1.5%甲醇诱导培养48h,表达产物抗菌活性最高。结论优化条件为HRP5′的大规模发酵奠定了基础。

【Abstract】 Objective To increase expression level,enhance antibacterial activity of expression products by optimizing expression conditions of recombinant Pichia Pastoris GS115-pPICZα-A-HRP5′.Methods Made some differences in the content of glycerol and the pH of yeast culture,the time before inducement and after inducement,and the induced content of methanol.Results Optimum expression conditions of HRP5 were obtained: content of glycerol was 2.0%,pH of yeast culture was 6.0,content of methanol was 1.5%,the time before and after inducement were 30hours and 48hours respectively.Conclusion Opitimized expression conditions made a solid foundation for the large scale formentation of HRP5′.
