小叶黑柴胡诱导人胃腺癌MGC-803细胞凋亡的实验研究
A Experimental Study of Bupleurum Smithii Wolff Var Parvifolium shan et Li on Inducing Apoptosis in Human Gastric Adenocarcinoma MGC-803 Cells
【摘要】 目的研究小叶黑柴胡诱导人胃腺癌MGC-803细胞的凋亡效应。方法实验分6组,对照组(MGC-803细胞培养于基础培养基中),实验1~5组(MGC-803细胞培养于基础培养基中加小叶黑柴胡的乙醚提取物,小叶黑柴胡终浓度分别为4、8、16、32、64μg/μL/3mL培养液)。计算各组细胞死亡率、细胞生长抑制率、克隆形成率并观察形态学变化,应用凝胶电泳、激光扫描共聚焦显微镜观察分析小叶黑柴胡作用MGC-803细胞后的细胞DNA含量的改变。结果小叶黑柴胡作用后细胞死亡率、细胞生长抑制率升高,克隆形成率降低,与对照组比较差异有统计学意义(P<0.01);细胞DNA含量荧光值测定,对照组(2265.92±225.00),实验组(1653.60±236.78),差异有统计学意义(P<0.01)。细胞DNA凝胶电泳实验组显示清晰的“阶梯状”条带。结论小叶黑柴胡可诱导人胃腺癌MGC-803细胞凋亡。
【Abstract】 Objective To study effects of apoptosis of Bupleurum smithii wolff Var Parvifolium shan et Li on human gastric adenocarcinoma MGC-803 cells.Methods Bupleurum smithii wolff Var Parvifolium shan et Liin different concentration(4、8、16、32、64μg/μL/3mL culture fluid) was administered to the MGC-803 cells in 6 groups.The changes of morphology、death rate、the inhibition rate of cell growth、 cloning efficiency were observed.DNA content of MGC-803 were measured with the laser scanning confocal microtechnic.Results Bupleurum smithii wolff Var Parvifolium shan et Li could significantly increase the death rate、the inhibition rate of cell growth.The cloning efficiency was reduced significantly(P<0.01).DNA content of MGC-803 in experimental group was significantly higher than that in controls.DNA gel eletrophoresis showed that a clear DNA ladder could be observed in experimental groups.Conclusion Bupleurum smithii wolff Var Parvifolium shan et Li could induce apoptosis in human gastric adenocarcinoma MGC-803 cells.
