药用植物白术DNA提取方法的研究
Study on Extracting Genomic DNA from Atractylodes macrocephala Koidz
【摘要】 为从白术叶片中提取高质量的基因组DNA,在传统CTAB法的基础上,采取先破碎细胞,将存在于细胞质中影响DNA提取质量的多酚等次生代谢物质去除后再裂解细胞核,经分离纯化,提取的DNA通过A260/A280比值测定,琼脂糖凝胶电泳和ISSR-PCR扩增检测,结果表明改良CTAB法提取的DNA纯度高,可作为ISSR分子标记的模板及用于后续分子生物学的研究。
【Abstract】 In order to get high-quality DNA from leaves of Atractylodes macrocephala Koidz,an efficient procedure was developed based on the conventional CTAB methods.The major improvement included elimination of secondary metabolites such as polyphenols and collection of nuclei before extracting DNA.The results from ratios of A260/A280,electrophoresis and ISSR-PCR amplification suggested that quality DNA extracted by using STAB methodproduces was higher than that extracted with conventional CTAB method.
