【摘要】 目的通过节律基因mPeriod1(mPer1)过表达,研究mPer1对小鼠Lewis肺癌细胞迁移的影响。方法采用脂质体介导法将mPer1基因转染入Lewis细胞。通过RT-PCR检测mPer1在Lewis细胞中的表达;采用Chemico公司QCM(tm)24-Well Colorimetric Cell Migration Assay检测细胞迁移的变化。结果与pcDNA3.1空质粒相比,pcDNA3.1-mPer1转染组mPer1表达增加,其迁移率与对照组比较明显下降。结论节律基因mPer1过表达能够抑制Lewis肺癌细胞的迁移。更多还原

【Abstract】 Objective To study the effects of circadian gene mPeriod1(mPer1) on migration of Lewis lung cancer cells by mPer1-overexpression.Methods Lipofectamine-mediated gene transfection method was used to transfect mPer1 gene into Lewis lung cancer cells.The expression of mPer1 gene was detected by RT-PCR and Western blotting.The cell migration was detected by Chemico QCM(tm) 24-Well Colorimetric Cell Migration Assay.Results As compared with the cells transfected with pcDNA3.1,the cells transfected with pcDNA3.1-mPer1 demonstrated the low migration.Conclusion The overexpressed mPer1 may inhibit the migration of Lewis lung carcinoma cells.更多还原
