不同镍含量金属植入对大鼠受孕和胚胎发育的影响

【摘要】 [目的]植入镍及镍钛合金于体内观察持续镍离子(Ni2+)释放对大鼠受孕和胚胎发育的影响。[方法]大鼠随机分为假手术组、镍组、合金组;植入镍及镍合金于股部,配对后观察对大鼠受孕和胚胎发育的影响。[结果]镍组的着床数、活胎率较假手术组和合金组低(P﹤0.05),死胎率高于假手术组和合金组(P﹤0.05),胎鼠的体重和肝、肺、肾脏器系数镍组低于假手术组和合金组(P﹤0.05),生后d1、5乳鼠体重镍组低于假手术组和合金组(P﹤0.05),10、15、20d乳鼠各组差异无统计学意义。[结论]金属镍植入物致大鼠着床数、活胎率下降,死胎率上升,子代体重和脏器系数降低,并影响子代的围生期存活能力,而镍钛合金相对具有较高的生物安全性。

【Abstract】 [Objective]To observe toxic effects of nickel ion releasing on pregnancy and embryogenesis after that nickel and nickel alloy were implanted into female rat hind limbs.[Methods]The rats were randomly divided into three groups:the sham operation group,nickel group and alloy group.Nickel(5 g/kg,n=15)and nickel alloy(10 g/kg,n=15)were respectively implanted into one hind limb contacting with thighbone in young Wistar female rats(75~80 day,180-200 g).Amphimixis was performed after 14 days,and then pregnancy and embryogenesis were studied.[Results]The rates of embryogenesis and nativity were significantly lower in nickel implanted rats than those of nickel alloy implanted rats and sham operated rats(P﹤0.05).The coefficients of weight,liver,lung and kidney of fetus declined respectively in nickel implanted rats(P﹤0.05)than those in nickel alloy implanted rats and sham operated rats.Neonatal rats in nickel alloy and sham operation groups were heavier than nickel group(P﹤0.05).[Conclusions]Implanted nickel causes toxic effects of reducing the ratio of embryogenesis and nativity of female rats,coefficients of weight organs of fetus and avoirdupois of neonatal rats.However,nickel alloy used in clinical treatment has favourable biologic safety.
