半对数回归在测定麻疹及风疹病毒抗体水平中的应用
APPLICATION OF SEMI-LOGARITHM REGRESSION IN DETERMINING THE ANTIBODY LEVEL
【摘要】 [目的]建立半对数回归ELISA法测定麻疹及风疹病毒的抗体水平。[方法]在常规ELISA法的基础上,以阳性参考血清的不同稀释度的对数为横坐标,与其492nm处的相应吸光度(OD值)进行线性回归,制定标准曲线。根据被测血清标本的吸光值推算其抗体滴度。[结果]统计学分析表明,抗体滴度的对数与其相应的吸光值呈显著线性相关(P﹤0.001)。用半对数回归推算的结果与常规ELISA法进行比较,差异无统计学意义(P﹥0.05)。[结论]该方法简便经济,用在进行大批量测定工作时,可节省大量的工作量和实验成本。
【Abstract】 [Objective]To establish a semi-logarithm regression method for determining the antibody titers of measles virus and rubella virus by ELISA in sera.[Methods]The logarithm of serial doubling dilution of the positive control serum was made as abscissa,semi-logarithm regression was made with their corresponding absorbances(OD values)in 492nm,and the antibody titers of serum specimens according to their absorbances in the standard curve was calculated.[Results]Statistics analysis indicated that the logarithm of antibody titers correlated well with their corresponding absorbances(P﹤0.001).Compared with the conventional ELISA,there was no significant difference(P﹥0.05).[Conclusions]The method is convenient and economical,especially in determining a large number of serum specimens.
