丙烯酰胺肝脏代谢与细胞色素P450 2E1蛋白表达
STUDY ON AA METABOLISM IN RAT LIVER AND THE EXPRESSION OF CYP2E1
【摘要】 [目的]了解丙烯酰胺(acrylamide,AA)在肝脏中的代谢机制。[方法]通过AA与大鼠肝切片共孵育,HPLC-MS/MS检测AA代谢性反应产物环丙氧酰胺(glycidamide,GA)的生成量,western blot分析细胞色素P4502E1(CYP2E1)蛋白的表达。[结果]0.5mmol/L剂量组中没有检测到GA,而其他3个剂量组(1.0、2.0、4.0mmol/L)则随着AA浓度的增加,肝切片代谢生成的GA也随之增加,3种剂量组间差异显著(P﹤0.05);相同的肝切片中由于AA浓度的不同,CYP2E1表达有所不同,而且随着AA浓度的增加其表达也增加,表明AA对CYP2E1的蛋白表达有诱导作用。[结论]AA可通过诱导使CYP2E1活性增加而增加GA的生成量,但低摄入量时AA详细的代谢机制及AA对CYP2E1的详细诱导机制还需要进一步研究。
【Abstract】 [Objective]To observe the metabolism of AA in rat liver.[Methods]AA was incubated with rat liver slices.The quantification of GA was determined by HPLC-MS/MS.The expression of CYP2E1 was analyed by western blot.[Results]GA was not detected in the liver slices which were incubated with AA in the dose of 0.5 mmol/L.For the other three doses(1.0、2.0、4.0 mmol/L),the concentration of GA increased corresponding to the increasing of AA doses,and there were significant differences of the GA concentrations among the three AA doses(P﹤0.05).The expression of CYP2E1 increased corresponding to the increasing of AA doses in the same slice.The inductivity of AA to CYP2E1 expression was obvious.[Conclusion]The concentration of GA increases by the enhance of CYP2E1 activity which is induced by the AA.However,the detailed mechanism of AA metabolism in the low dose and the inductivity of AA to CYP2E1 expression are still unclear.
