GSTM1基因多态性与肺癌易感性的病例对照研究
A CASE-CONTROL STUDY OF THE IMPACT OF GLUTATHIONE S-TRANSFERASE M1 POLYMORPHISM ON THE RISK OF LUNG CANCER
【摘要】 [目的]探讨谷胱甘肽硫转移酶μ基因(GSTM1)缺失和吸烟、饮酒与肺癌易感性的关系。[方法]采用病例对照研究方法,调查42例病例和103例对照的吸烟、饮酒习惯,应用PCR技术检测GSTM1的基因型。采用lo-gistic回归分析GSTM1基因型与肺癌的关系及其与吸烟量、饮酒的交互作用。[结果]病例组GSTM1缺失率57.1%,对照组55.3%,两组差异无统计学意义,经调整混杂因素后,GSTM1缺失联合重度吸烟(累计吸烟量≥30包×年)的OR值12.841(2.322~71.008),交互作用系数为2.796。GSTM1联合饮酒的OR值10.728(1.270~90.656),交互作用系数为1.651。[结论]GSTM1基因缺失与重度吸烟(累计吸烟量≥30包×年),GSTM1基因缺失与饮酒均对肺癌的发生具有交互作用。
【Abstract】 [Objective]To investigate lung cancer patients’exposure risk of the Glutathione S-transferase M1 deletion(GSTM1-null),smoking and alcohol drinking,a case-control study was conducted.[Methods]42 cases with lung cancer and 103 healthy controls were recruited for the study.Logistic regression analysis was performed to calculate the odds ratio(OR)and OR 95%confidence intervals(CI)of GSTM1-null genotype,smoking and alcohol drinking.Meanwhile,the inter-action between GSTM1-null genotype and smoking and between GSTM1-null and drinking were estimated by interaction coeffi-cient(γ).[Results]The results showed that the prevalence of the GSTM1-null genotype in the cases and controls were not significantly different,with frequency of 57.1%and 55.3%respectively.Compared to non-smokers or light smokers(﹤30 pack-years)with at least one positive GSTM1 genotype,Heavy smokers(≥30 pack-years)with GSTM1-null genotype showed a increased risk for lung cancer,with the OR=12.841(95%CI:2.322~90.656),and interaction coefficient was 2.796.Alcohol drinkers carrying GSTM1-null genotype also had a higher risk to lung cancer(OR=10.728;CI,1.270~ 90.656)than non-drinkers with at least one positive GSTM1gene,and interaction coefficient of alcohol drinking and GSTM1-null was 1.651.[Conclusion]Our study suggests that the GSTM1-null genotype may have interaction with smoking and dink-ing for lung cancer.
