mda-7/IL-24基因对卵巢癌细胞耐药性的影响
Effects of mda-7/IL-24 gene on the drug-resistance of ovarian cancer cell lines.
【摘要】 目的:探讨抑癌基因mda-7/IL-24对卵巢癌细胞株A2780和耐药株A2780/ DDP耐药性的影响。方法:构建人mda-7/IL-24基因重组腺病毒,感染A2780和A2780/ DDP的两种卵巢癌细胞株,MTT法检测细胞感染对顺铂(DDP),阿霉素(ADM),5-氟尿嘧啶(5-FU),泰素(Taxol)4种化疗药物敏感性的变化。结果:重组腺病毒载体Ad.mda-7/ IL-24经测序、酶切电泳检测正确;感染后A2780细胞对DDP、5-FU、Taxol的耐药性降低, DDP的药物敏感性提高了15.45倍,5-FU和Taxol的药物敏感性分别提高了2.15倍和3.63倍;A2780/DDP细胞对5-FU药物敏感性提高了2.62倍。结论:成功构建了Ad.mda-7/IL-24重组腺病毒载体,感染卵巢癌细胞A2780可提高对DDP、5-FU和Taxol的敏感性。
【Abstract】 Objective:To study the effects of tumor suppressor gene mda-7/IL-24 on the ovarian cancer cell line A2780 and drug-resistant cell line A2780/DDP.Methods:Adenovi- ral-mediated mda-7/IL-24 (Ad.mda-7/IL-24) was constructed and used to infect two cell lines.Cells sensitivity to four chemotherapeutic drugs,including DDP,ADM,5-FU and Taxol, were examined with methyl thiazolyl tetrazolium (MTT) rapid photocolorimetric assay.Results: The recombinant Ad.mda-7/IL-24 was conformed by sequence analysis and electrophoresis.Af- ter Ad.mda-7/IL-24 transfection,A2780 drug-resistance to DDP,5-FU and Taxol of A2780 was decreased.Sensitivity to DDP of A2780 was increased by 15.45 times,and sensitivity to 5-FU and Taxol was increased by 2.15 and 3.63 times,respectively.Infected A2780/DDP cells had higher sensitivity to 5-FU,which increased 2.62 times.Conclusions:The recombinant Ad.mda- 7/IL-24 is successfully constructed,mda-7/IL-24 gene transfection is capable of increasing the sensitivity of A2780 cells to DDP,5-FU and taxol.
