中国荷斯坦牛微卫星基因座遗传多态性研究
Polymorphism distributions of microsatellite loci in Chinese Holstein cattle
【摘要】 利用PCR技术和复合电泳银染技术检测了中国荷斯坦牛ETH225、BM1824、BM2113和ETH152 4个微卫星座位的多态性分布,计算了各个基因座的杂合度(H)、有效等位基因数(Ne)、Shannon信息熵(S)、多态信息含量(PIC)和固定指数(F)。结果显示,4个微卫星基因座的基因型分布均不符合Hardy-Weinberg平衡,其中,基因座ETH225的H、Ne、S、PIC、F均为最高,分别为0.913 0,11.488 2,2.574 4,0.906 6和-0.095 3,表明该座位的遗传多态性比其他座位丰富,具有更大的选择潜力。
【Abstract】 The polymorphism distributions of microsatellite loci,ETH225,BM1824,BM2113 and ETH152 were detected in Chinese Holstein cattle by Polymerase Chain Reaction and multiplex gel electrophoresis followed by silver staining.Heterozygosity(H),effective number of alleles(Ne),Shannon’s information index(S),polymorphism information content(PIC) and fixation index(F) were calculated.Results showed that all loci did not obey Hardy-Weinberg equilibrium.H,Ne,S,PIC and F of ETH225 locus were the highest,and they were 0.913 0,11.488 2,2.574 4,0.906 6,-0.095 3,respectively,which showed that the polymorphism of ETH225 locus was richer than that of the others and this locus had more possibilities to be chosen.
