核因子NF-κB对气道平滑肌细胞增殖的影响
Effects of nuclear factor kappa-B(NF-κB) on proliferation of airway smooth muscle cells
【摘要】 目的:研究核因子-κB(NF-κB)对大鼠气道平滑肌细胞(ASMC)增殖活性的影响。方法:体外培养ASMCs,采用EMSA法直接检测ASMC中NF-κB的活性,MTT法、细胞周期检测及细胞核增殖抗原(PCNA)的免疫细胞化学染色法观察细胞增殖情况。结果:血小板活化因子刺激后ASMC中NF-κB明显活化,各细胞增殖指标均显著增高(P<0.05),NAC可显著抑制上述效应(P<0.05)。结论:NF-κB的活化能直接参与ASMC的增殖。
【Abstract】 Objective:To investigate the effect of nuclear factor-kappa B(NF-κB) on the proliferation of airway smooth muscle cells(ASMC).Methods: NF-κB activity in ASMC was examined by electrophoretic mobility shift assay(EMSA).MTT,flow cytometry and immunohistochemistry for proliferating cell nuclear antigen(PCNA) were used to evaluate ASMC proliferation.Results: Platelet-activating factor(PAF) stimulated the NF-κB activity and the cell proliferation in ASMC.The positive rate of PCNA was significantly different among these groups.N-acety-L-cysteine(NAC) markedly inhibited the increased NF-κB activity and ASMC proliferation induced by PAF.Conclusion: NF-κB activation may directly involve in the proliferation of ASMC,suggesting its important role in airway remodeling.
