应用微量滴定法测定天花疫苗效力
Detection of the potency of smallpox vaccine by microtitration
【摘要】 微量滴定法代替血球吸附法测定天花疫苗效力。通过对Vero细胞接种浓度和细胞病变判定时间的优化,确定了微量滴定法测定天花疫苗效力的方法,用Vero细胞微量滴定法和血球吸附法测定14批天花疫苗。两种滴定方法的检测结果差异有显著意义(P<0.05),二者存在正相关(r=0.76,0.001<P<0.05)和直线回归(b=0.45,0.001<P<0.05)关系,直线回归方程为y^=4.61+0.45x。微量滴定法变异系数CV为1.25%-2.89%。因此,微量滴定法敏感、可靠、操作简单、重复性好,可以代替血球吸附法。
【Abstract】 The titration of potency of smallpox vaccine by microtitration instead of hemadsorption was carried out in this study.Microtitration the potency of smallpox vaccine was set up by selection of inoculated Vero cell concentration and the time in calculation of the virus titration.Fourteen lots of smallpox vaccine were titrated by microtitration on Vero cells and hemadsorption on CEFs.The difference is significant between the results of the two methods (P<0.05).There are positive correlation(r=0.76,0.001<P<0.05),and linear regression relationship(b=0.45,0.001<P<0.05) between them,=4.61+0.45x.CV of microtitration is in a range 1.25%~2.89%.Therefore,microtitration can replace hemadsorption,because its sensitivity,reliability,and repetition are better than those of hemadsorption,and it is easy to operate.
