胰腺癌bcl-xL、cyclin D1的表达及其与STAT3的相互关系
Expression of bcl-xL and cyclin D1 in pancreatic carcinoma and their relationship with signal transducer and activator of transcription-3(STAT3)
【摘要】 目的:探讨凋亡抑制基因bcl-xL、细胞周期蛋白(cyclin)D1在胰腺癌组织中的表达情况及其与活化的信号传导和转录激活因子-3(phosphorylated signal transducer and activator of transcription-3,PSTAT3)、胰腺癌临床病理特征的相互关系。方法:采用免疫组织化学检测了41例胰腺癌组织和20例正常胰腺组织中PSTAT3、bcl-xL、cvclin D1、Ki67的蛋白表达,用TUNEL法检测41例胰腺癌组织中的凋亡指数(AI)。结果:胰腺癌组织中PSTAT3、bcl-xL和cyclin D1蛋白的表达分别为70.7%(29例)、87.8%(36例)、61.0%(25例),明显高于正常胰腺组织。bcl-xL、cyclin D1蛋白的表达与PSTAT3的表达均呈正相关(P<0.05),PSTAT3的表达与胰腺癌的临床分期、淋巴结转移、肿瘤细胞增殖指数(PI)、AI密切相关(P<0.05)。结论:胰腺癌组织中bcl-xL、cyclin D1的过度表达与PSTAT3密切相关,STAT3信号通路在胰腺癌的增殖凋亡过程中起重要作用。
【Abstract】 Objective To investigate the expression of anti-apoptosis genes bcl-xL and cyclin D1 in human pan- creatic carcinoma tissues,and their relationship with clinico-pathological characteristics including the expression of phos- phorylated signal transducer and activator of transcription-3 (PSTAT3).Methods PSTAT3,bcl-xL and cyclin D1 protein expression in 41 pancreatic cancer tissues and 20 normal pancreatic tissues were detected by immunohistochemical stai- ning.Simultaneously,the proliferative index (PI) and apoptotic index (AI) of the tumor cells were scored by Ki67 and TUNEL staining,respectively.Results The expression rates of PSTAT3,bcl-xL and cyclin D1 in pancreatic carcinoma were 70.7%(29/41),87.8%(36/41) and 61.0%(25/41),respectively,significantly higher than those in the normal pancreatic tis- sues.Positive correlation was found between the expression of bcl-xL,cyclin D1 and PSTAT3 (P<0.05).Furthermore, PSTAT3 expression was closely related with the clinical staging,status of lymph node metastasis,increased PI and decreased AI(P<0.05).Conclusions Activated STAT3 is closely related to the over-expression of anti-apoptosis genes bcl-xL and cy- clin D1 in human pancreatic carcinoma,and might play an important role in the tumorigenesis of this lethal malignancy.
