【摘要】 目的研究磷脂酰激酶3通路抑制剂渥曼青霉素(wortmannin)对聚腺苷二磷酸核糖聚合酶(PARP)蛋白表达的影响。方法应用DNA琼脂糖凝胶电泳检测胃癌细胞BGC-823凋亡,以蛋白印迹法检测PARP蛋白及活化片断表达情况。结果足叶乙甙(etoposide)及阿霉素(doxorubicin)能诱导胃癌细胞凋亡及BGC-823 PARP裂解的发生,wortmannin可以增强其作用。结论磷脂酰激酶3通路抑制剂渥曼青霉素可以增强胃癌细胞凋亡的发生。更多还原

【Abstract】 Objective To investigate the effect of wortmannin,the inhibitor of phosphatidylinositol 3′-kinase path,on the expression of the poly ADP-ribose polymerase protein(PARP). Methods The agarose gel electrophoresis of DNA was used to detected apoptosis of gastric carcinoma cells BGC-823,and the expression of the poly ADP-ribose polymerase protein and its activated fragment were detected by Western-blotting assays. Results Etoposide and doxorubicin could induce gastric carcinoma cell apoptosis and split of the poly ADP-ribose polymerase protein in BGC-823 cells,wortmannin could enhance the effect. Conclusion The inhibitor of phosphatidylinositol 3′-kinase path could up-regulate the apoptosis of gastric cancer cells.更多还原
