【摘要】 [目的]以血清药理学的方法观察血化瘀通脉注射液(HYTM)对于培养人脐静脉内皮细胞ECV304增殖的影响,为治疗性血管新生的靶基因选择提供线索。[方法]取对数生长期的内皮细胞,同步化于G0期,2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐(CCK-8)检测细胞增殖活性;酶联免疫吸附测定(ELISA)法检测5溴-2脱氧尿苷(BrdU)的掺入。[结果]与空白血清对照组相比,10%的HYTM含药血清体积添加分数可显著提高细胞增殖活性。[结论]10%的HYTM含药血清体积添加分数通过促进细胞DNA合成,可能具有保护血管内皮的作用。更多还原

【Abstract】 [Objective] To observe the influence of Hua Yu Tong Mai (Removing stasis and promoting circulation , HYTM) Injection on the proliferation of ECV304 in human umbilical vein with the serum pharmacological method and provide a clue for choosing target gene for promoting vascular angiogenesis. [Methods] The endothelial cell in logarithmic growth phase was obtained and synchronized in the G0 time. Their proliferation activity was examined by CCK-8; BrdU incorporate was examined with ELISA. [Results] Compared with the blank blood serum control group, the volume increase score of medicine-containing serum with 10% HYTM. [Conclusions] The volume increase score of serum containing medicine of 10% HYTM may promote endothelial cell proliferation through promoting DNA synthesis and mitosis and possibly has protective effect on vascular endothelium.更多还原
