转导血管内皮生长因子预防血管成形术后再狭窄的实验研究
Experimental Study on Vascular Endothelial Growth Factor Transduction in Prevention of Restenosis after Angioplasty
【摘要】 目的探讨血管内皮生长因子(VEGF)预防血管成形术后再狭窄。方法建立动脉粥样硬化兔模型,利用球囊在该模型腹主动脉导入pcDNA3.0/hVEGF165(n=10),对照组导入空载体(n=10)。采用RT-PCR法检测外源VEGF基因的表达。术后2、4周观察转导外源基因对血管内皮修复的影响。术后1、2、4周行MRI检查,观察左肾动脉开口处上段被扩张腹主动脉的管腔面积变化。结果利用球囊导管能成功转导pcDNA3.0/hVEGF165,治疗组和对照组完成内皮修复的时间分别为2周和4周,术后2周治疗组和对照组的管腔面积分别为(7.95±1.42)mm2和(6.74±1.01)mm2,术后4周分别为(7.40±2.29)mm2和(2.21±1.44)mm2。结论在动脉粥样硬化兔模型局部血管利用球囊导管能成功地转导VEGF基因,有效促进局部血管内皮细胞的生长,取得抑制局部血管管腔狭窄的作用,这对于防止血管介入术后再狭窄发生具有十分重要的意义。
【Abstract】 Objective To evaluate hVEGF165 transduction in prevention of restenosis after angioplasty.Methods Rabbit models with atherosclerosis were established.The pcDNA3.0 recombinant hVEGF165 was transferred to ventral aorta by Foley’s tubes,and the pcDNA3.0 was transferred in the controls.The expression of hVEGF165 was observed by RT-PCR.The pathological specimens were obtained by killing the rabbits on the week 2 and 4 after hVEGF165 gene transfer.The pathological specimens were evaluated by computerized image analysis system.The areas of ventral aortic lumina were measured by MRI after angioplasty.Results The pcDNA3.0/ hVEGF165 was transferred successfully by Foley’s tube.The restoring time of endothelium was 2 weeks in the treated group and 4 weeks in the control group,respectively.The area of canal in the treated group and the control group was(7.95±1.42) mm2 and(6.74±1.01) mm2 after 2 weeks,(7.40±2.29) mm2 and(2.21±1.44) mm2 after 4 weeks,respectively.Conclusion hVEGF165 gene transfer could be achieved by Foley’s tube.It could accelerate the restoration of endothelial integrity and inhibit the restenosis in the atherosclerosis arteries after hVEGF165 gene transfer. The procedure could be useful in preventing restenosis after angioplasty.
