成纤维细胞与大肠癌细胞相互作用对大肠癌细胞表达EMMPRIN的影响
Effects of fibroblasts-colorectal cancer cells interaction on EMMPRIN expression in colorectal cancer cells
【摘要】 目的研究成纤维细胞与大肠癌细胞相互作用对大肠癌细胞表达细胞外基质金属蛋白酶诱导因子(extracellu-lar matrix metalloproteinase inducer,EMMPRIN)的影响。方法建立大肠癌细胞SW620与HELF成纤维细胞共培养模型,并设SW620细胞单独培养为对照组,通过RT-PCR和免疫细胞化学方法检测对照组和共同培养组SW620细胞中EMMPRIN的表达。结果共同培养组SW620细胞EMMPRIN的表达在mRNA水平和蛋白水平均高于对照组。结论成纤维细胞与大肠癌细胞相互作用促进了大肠癌细胞EMMPRIN表达增加,EMMPRIN表达增加可能在大肠癌浸润和转移中发挥重要作用。
【Abstract】 Objective To research the effects of fibroblasts-colorectal cancer cells interaction on extracellular matrix metalloproteinase inducer(EMMPRIN) expression.Methods Co-cultured model of fibroblasts HELF and colorectal cancer cells SW620 was established.Colorectal cancer cells were lonely cultured as control.The expression of EMMPRIN in lonely cultured and co-cultured colorectal cancer cells were detected by RT-PCR and immunohistochemical staining.Results EMMPRIN expression in co-cultured group was higher than that in lonely cultured group significantly,both at the levels of mRNA and protein.Conclusion Interaction between colorectal cancer cells and fibroblasts can promote EMMPRIN expression,which may play an important role in colorectal cancer invasion and metastasis.
