葡聚糖硫酸酯的超滤分级及其抗凝血活性研究

【摘要】 目的研究各种葡聚糖硫酸酯(GS)超滤分级的体外和对大鼠的体内抗凝血活性。方法通过超滤分级的方法获得截留分子量不同的GS级分,并测其特性黏度[η];分别研究不同[η]的GS对大鼠凝血时间(CT)和全血复钙时间(RT)的时效性和量效性。结果各种[η]的GS均能显著延长CT和RT,且呈剂量依赖性;GS给药2 h时抗凝血活性达到最大;不同[η]的GS对CT和RT的延长作用不同,[η]为274.8 m l/g的GS的抗凝血活性最强。结论不同[η]GS均具有抗凝血活性,[η]为274.8 m l/g的GS抗凝血活性最强,与活性为175 IU/mg的低分子量肝素相当。

【Abstract】 Objective To assess the anticoagulative activity of glucosan sulfate with different intrinsic viscosity in vivo and in vitro.Methods Glucosan sulfates with different intrinsic viscosity were obtained by multiple ultra-filtration of glucosan sulfate raw material and their intrinsic viscosities were measured.Coagulation time(CT) and recalcification time(RT) were measured using a test tube method.Results CT and RT increased by the anticoagulation of fractioned glucosan sulfates,and they showed a significant dose-dependent relationship.The anticoagulation effect was maximum after 2 h of administration.The CT and RT were different with different fractions of glucosan sulfates.The anticoagulation of the glucosan sulfate with intrinsic viscosity of 274.8 ml/g was the highest.Conclusion The anticoagulation can be enhanced significantly by the glucosan sulfate with different intrinsic viscosity.The higher the intrinsic viscosity was,the higher the effect of anticoagulant activity was.The anticoagulant activity of the glucosan sulfate fraction with an intrinsic viscosity of 274.8 ml/g was the same as 175 IU/mg of low molecular weight heparin.
