CDX2、PTEN在结直肠癌中的表达及其与ki67相关分析
Expression of CDX2、PTEN and correlation to ki67 in colorectal carcinoma
【摘要】 目的探讨CDX2、PTEN在结直肠癌中的表达及其与ki67相互关系。方法采用免疫组织化学方法检测人50例结直肠癌组织中三者的表达情况。结果CDX2、PTEN与肿瘤的TNM分期,组织分级,淋巴结转移,原发灶浸润程度有关。Pearson积差相关系数r值为0.406。二者与ki67呈负相关(r值分别为-0.684和-0.511)。三者联合分析显示:CDX2(+)PTEN(+)组的ki67平均标记指数显著低于单独CDX2(+)或PTEN(+)组,或共同表达阴性组。结论在调节肠上皮细胞增殖分化方面PTEN对CDX2有调控作用。CDX2、PTEN及ki67在结直肠癌发生、发展中起着重要作用,三者联合检测有助于结直肠癌细胞增殖活性,恶性程度的判断。
【Abstract】 Objective To investigate the expression of CDX2、PTEN and ki67 in colorectal cancer,and the correlations among CDX2、PTEN and ki67. Methods Fifty samples of human colorectal carcinoma were detected for CDX2、PTEN as well as ki67 by immunohistochmistry. Results The expression of CDX2 and PTEN in colorectal carcinoma have both related with tumors TNM stages、tissue grading、lymph-node metastasis and primary focus infiltrating depth. Coefficient of product-moment correlation is 0.406. CDX2 and PTEN expressions were negatively correlated with the expression of ki67 r=-0.684,P<0.000; r=-0.511, (P<0.000) respectively. Analysis of CDX2、PTEN and ki67 showed that the ki67 average labeling index of CDX2(+)PTEN(+) group was significantly lower than that of CDX2(+) group alone、PTEN(+)group alone or both negative group. Conclusion CDX2, intestinal-specific homeobox genes, regulates cell proliferation and differentiation in the continuously renewed intestinal epithelium. PTEN positively regulates CDX2 gene expression; The positive expression of CDX2、PTEN and ki67 may play an important role in the development and progression of colorectal carcinoma. Combined detection of 3 indices may be helpful for the cell proliferation and malignant degree.
