茶树油纳米脂质体制备及处方设计与优化
Optimization of the preparation and prescription of nano-sized tea tree oil liposomes
【摘要】 目的优化制备茶树油纳米脂质体的处方。方法采用乙醇滴入-超声工艺制备茶树油纳米脂质体;并以茶树油脂质体的包封率为主要评价指标,采用正交设计法优化茶树油脂质体的配方;综合考虑茶树油脂质体的粒径及载药量2个因素,确定茶树油脂质体的处方。结果获得了茶树油脂质体的工艺处方:卵磷脂质量浓度为10 g.L-1、卵磷脂与胆固醇的质量比为5∶1、茶树油质量浓度为1.0 g.L-1、VE的质量浓度为1.0 g.L-1、去氧胆酸钠的质量浓度为0.5 g.L-1、吐温-80的质量浓度为4.0 g.L-1,水相介质是pH值为6.8的PBS缓冲溶液。按该处方工艺组合制备3批茶树油脂质体,包封率平均值为(67.34±1.06)%,粒径为(65.0±30.3)nm。结论按照优化处方,采用乙醇滴入-超声工艺可制得的包封率稳定、粒径较小且分布较窄的茶树油纳米脂质体。
【Abstract】 Objective To optimize the prescription of tea tree oil liposomes.Methods The liposomes were prepared by the ethanol injection-ultrasonic method;The envelope efficiency of tea tree oil liposomes was taken as the criteria,the prescription of the liposomes were optimized by orthogonal design.And the diameter and the quantity of the tea tree oil that wrapped in the liposomes were taken into account as the subordinate criterias.Results The best prescription consisted of egg phospholipids(10 g·L-1),egg phospholipidscholesterol(m:m=5∶1),concentration of tea tree oil(1 g·L-1),concentration of VE(1.0 g·L-1),concentration of sodium deoxycholate(0.5 g·L-1),concentration of Tween-80(4.0 g·L-1),the water phase phosphate buffer solution(pH 6.8).According to the prescription,the liposomes had homogeneous shape with the particle size of(65.0±30.3)nm and the envelope efficiency of(67.34±1.06)%.Conclusions The optimized formulation of tea tree oil liposomes was reasonable,practicable with high envelope efficiency and low particle size.
