RP-HPLC法测定蒙药森登-4汤中栀子苷、槲皮素和没食子酸的含量

【摘要】 目的建立测定蒙药森登-4汤中栀子苷、槲皮素和没食子酸的高效液相色谱方法。方法栀子苷的测定条件为:Hypersil C18 ODS(250.0 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-水(体积比为13.5∶86.5),流速为0.8 mL.min-1,检测波长为238 nm。槲皮素的测定条件为:Century SIL C18 BDS(250.0 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-水-磷酸(体积比为60.00∶40.00∶0.02),流速为0.8 mL.min-1,检测波长为360 nm;没食子酸的测定条件为:Century SIL C18 BDS(250.0 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-水-磷酸(体积比为5.00∶95.00∶0.02),流速为0.80 mL.min-1,检测波长为272 nm。结果在各自的色谱条件下,栀子苷、槲皮素和没食子酸质量浓度与峰面积具有良好的线性关系。平均回收质量分数分别为:栀子苷96.9%(RSD=1.9%,n=6);槲皮素97.1%(RSD=2.3%,n=6);没食子酸98.7%(RSD=1.7%,n=6)。结论该方法可作为该制剂的含量测定方法。

【Abstract】 Objective To establish the HPLC method for determination of geniposide,quercetin and gallic acid in Mongolian medicine Sendeng-4 decoction.Method The determination of geniposide was performed with column Hypersil ODS C18(250 mm×4.6 mm,5 μm) using acetonitrile-water(V∶V=13.5∶86.5) as mobile phase at a flow rate of 0.8 mL·min-1.UV detective wavelength was set at 238 nm.The determination of quercitin was performed on Century SIL C18 BDS(250 mm×4.6 mm,5 μm)column,using methanol-water-phosphoric acid(V∶V∶V=60.00∶40.00∶0.02) as mobile phase at a flow rate of 0.8 mL·min-1 and UV detection at 360 nm.The determination of gallic acid was performed on Century SIL C18 BDS(250 mm×4.6 mm,5 μm) column,using acetonitrile-water-phosphoric acid(V∶V∶V=5.00∶95.00∶0.02) as mobile phase at a flow rate of 0.8 mL·min-1 and UV detection at 272 nm.Results The standard curves of geniposide,quercetin,and gallic acid had a good linearity under the HPLC conditions.The average recovery and relative deviation were 97.1% with 2.3% for geniposide,96.9% with 1.9% for quercetin,and 98.7% with 1.7% for gallic acid,respectively.Conclusion The methods are simple,accurate,and can be used as methods for the determination of Sendeng-4 decoction.
