钙调素在格列美脲促进正常大鼠骨骼肌细胞糖原合成中的作用
The Effect of Calmodulin on Glycogen Synthesis in Glimepiride-Treated Skeletal Muscle in Normal Rats
【摘要】 目的探讨钙调素(CaM)在格列美脲(GM)促进正常大鼠骨骼肌糖原合成中的作用。方法分离大鼠骨骼肌并进行体外培养,将细胞分成含胰岛素和不含胰岛素两大组,每组又分成含CaM拮抗剂R24571和不含R24571两亚组,各亚组分别设对照,给予不同浓度GM(0.01、0.10、1.00、10.00μmol/L)孵育4h后进行细胞CaM活性和糖原含量测定。结果在胰岛素存在下,加入GM后,CaM活性和糖原含量都明显升高(P<0.05),且在一定浓度范围内随着GM浓度升高而升高;加入R24571后,CaM活性明显受抑,糖原含量明显下降(P<0.05)。在无胰岛素存在下,加入GM后,CaM活性明显升高且在一定范围内随GM浓度升高而升高,糖原含量无明显变化(P>0.05)。结论CaM在GM促进胰岛素介导的骨骼肌糖原合成中起重要作用。
【Abstract】 Objective To investigate the effect of calmodulin(CaM)on the glycogen synthesis in glimepiride(GM)-treated skeletal muscle cells in normal rats.Methods The skeletal muscle cells of normal rats were cultured in vitro.Based on the existing of insulin,the cells were grouped into two,each with two subgroups:with or without the specific inhibitor of CaM R24571.In each subgroup,the cells were cultured at different concentration of GM(0.01,0.10,1.00,10.00 μmol/L)for 4 hours.The cells were then collected for measuring the activity of CaM and the content of glycogen.Results With the presence of insulin,GM enhanced the activity of CaM and the content of glycogen in a dose-dependent fashion(P< 0.05).After R24571 was added,the activity of CaM was inhibited and the content of glycogen declined obviously(P< 0.05).On the contrary,the activity of CaM rised while the content of glycogen had no obvious changes with the absence of insulin in the medium(P >0.05).Conclusion CaM plays a key role in GM involvement in insulin-induced glycogen synthesis.
