人脐带间充质干细胞向心肌样细胞分化的研究

【摘要】 目的探讨人脐带间充质干细胞(MSCs)向心肌样细胞分化可能性,为心肌细胞移植探索新细胞来源。方法采用5-氮杂胞苷(5-Aza)和二甲基亚砜(DMSO)诱导人脐带间充质干细胞分化。观察诱导后分化细胞形态变化,用免疫组织化学方法检测分化心肌样细胞的心肌肌钙蛋白I(troponin I)、心肌肌钙蛋白T(troponin T)和心肌结蛋白(desmin),逆转录聚合酶链反应(RT-PCR)测定分化的心肌样细胞是否有细胞心肌特异转录因子(Nkx2.5)和心肌细胞desmin的cDNA表达。结果人脐带MSCs经5-Aza和DMSO诱导后可向心肌样细胞分化,分化细胞表达心肌细胞的标记troponinI、troponinT和desmin。RT-PCR检测证实,人脐带MSCs诱导前不表达Nkx2.5和desmin,经5-Aza和DMSO诱导后表达心肌细胞标记Nkx2.5和desmin。结论人脐带间充质干细胞可以分化为心肌样细胞,5-Aza和DMSO可作为心肌细胞诱导剂,人脐带间充质干细胞可以作为心肌细胞的来源。

【Abstract】 Objective To investigate human umbilical cord mesenchymal stem cells(MSCs)differentiated into cardiomyocytes,and find a new source of cells for transplantation of cardiomyocytes.Methods MSCs were induced by incubated 5-azacytidine(5-Aza)and dimethylsulfoxide(DMSO).The myogenic cells differentiated were then evaluated by immunohistochemistry stained for desmin,cardiac troponin I and cardiac troponin T.The method of RT-PCR was used to test if the cells induced could express Nkx2.5 and desmin cDNA.This new cell line provided a powerful model for the study of cardiomyocyte differentiation.Results The MSCs derived from human umbilical cord could differentiate into cardiomyocytes induced by both 5-Aza and DMSO.Immunocytochemistry showed that differentiated cardiomyocyte-like cells expressed both desmin,cardiac troponin I and cardiac troponin T positive.The cells expressed Nkx2.5 and desmin cDNA after induced.Conclusions The MSCs derived from human umbilical cord Wharton’s Jelly can differentiate into cardiomyocytes induced by both 5-Aza and DMSO.Based on these observations,and taking into account immense advantages of UCB over than other sources of stem cell,human umbilical cord-derived MSCs retain myocardiogenic potential suitable for cell therapy and transplantation against intractable heart diseases.
