壳聚糖酶解液对抗药性大肠杆菌的抑制作用
Inhibition on Anti-ampicillin E.coli Strain by Enzymatic Solutions of Chitosan
【摘要】 采用微波法从根霉菌丝体中提取壳聚糖,经纤维素酶非特异性降解得到其寡糖溶液。采用其中初步抑菌效果较好的COS3-2、COS4-2、COS4-3、COS4-4分别对氨苄青霉素抗性大肠杆菌进行抑菌实验,结果表明:壳聚糖纤维素酶解液COS3-2、COS4-2、COS4-3、COS4-4对抗药性大肠杆菌均有较好的抑制作用,其中酶解液COS3-2的抑菌活性最强,当浓度为2.5%时抑菌圈直径达到18.6 mm,最低抑菌浓度(minimal inhibiting concentration,MIC)为0.05%;壳聚糖酶解液的抑菌作用随着酶解液浓度的增加而增强,并且抑菌作用需要一定的环境,在pH值5.5~6.0范围内,对氨苄青霉素抗性大肠杆菌有很好地抑制作用,pH值过高抑菌作用减弱。
【Abstract】 Chitosan was obtained from Rhizopus oryzae by microwave extracting method,and chose its cellulase solutions COS3-2、COS4-2、COS4-3、COS4-4 were used to do antimicrobial trial.The results suggested that COS3-2,COS4-2,COS4-3 and COS4-4 all had certain suppression.COS3-2 had the strongest suppression,inhibiting diameter over 18.6mm with 2.5% concentration.Minimal inhibiting concentration(MIC) was 0.05%.The higher concentration,the stronger suppression was.They restrained drug resistant E.coli well during pH5.5~6.0,while its suppressing ability would weak if pH was too high.
