利用猪子宫三种体外培养细胞构建三维医学模型
Establishment of in vitro three-dimensional medicine model with three types of swine uterus cell culture
【摘要】 目的在体外培养出结构和功能与活体组织和器官相同或相似的结构功能体,以作为器官移植宝贵材料或其他生物现象的体外研究模型。方法运用胰蛋白酶消化、细胞筛网法和组织块贴壁法分离构建三维医学模型所需的种子细胞,然后选择适宜的生物支架材料,最后将种子细胞依照自然子宫结构附植在支架材料上,经体外培养而获得的与自然子宫形态与功能相似的模拟体。结果本实验成功地建立了上皮细胞、基质细胞和平滑肌细胞的有限细胞系,并构建了猪子宫三种体外培养细胞三维医学模型。结论运用胰蛋白酶消化、细胞筛网法和组织块贴壁法分离种子细胞是一种较好的方法,其分离结果细胞较纯。
【Abstract】 Objective To construct a tissue-like culture in vitro for biological research.Methods Trypsin digestion and tissue adherent cell cultivation were used to isolate cells and uterus-like model was then constructed with the cells isolated and appropriated biological materials.Results Swine epithelium cell,stromal cell and smooth muscle cell lines were successfully established.The in vitro uterus-like three dimensional model was constructed.Conclusion Trypsin digestion and tissue adherent cell cultivation are good methods to separate seed cells.
