热休克蛋白70反义寡核苷酸诱导胶质细胞瘤凋亡的研究
The study of HSP70 antisense oligomer inducing apoptisis of neuropongioma cells
【摘要】 目的:探讨热休克蛋白70(HSP70)反义寡核苷酸阻断胶质瘤细胞的HSP70的表达及对胶质瘤细胞凋亡的影响。方法:用MTT法、流式细胞术、琼脂糖凝胶DNA电泳方法观察HSP70反义寡核苷酸对体外培养的胶质瘤C6细胞株HSP70表达的抑制及其对生长和凋亡的影响。结果:用HSP70反义寡核苷酸处理的胶质瘤细胞表现出明显的生长抑制,琼脂糖凝胶电泳显示凋亡细胞DNA降解的特征性DNA阶梯图;MTT法测定的生长抑制率和流式细胞术分析的凋亡率与HSP70反义寡核苷酸呈剂量、时间依赖性关系。结论:HSP70反义寡核苷酸通过抑制HSP70表达而诱导胶质瘤C6细胞株凋亡并抑制其生长,可作为基因治疗胶质瘤的新靶向。
【Abstract】 Objective:To investigate the influence of expression of HSP70 and apoptosis of tumor cell with HSP70 antiense oligomer in neuropongioma C6 cells.Methods:The effects of HSP70 antisense oligomer on HSP70 expression levels,the proliferation and apoptosis of neuropongioma C6 cells in vitro were studied by MTT test,flow cytometry and agarose gel DNA electrophoresis.Results:The inhibiting proliferation rate by MTT test and the induced apoptosis rate by flow cytometry analyses revealed the apoptosis and inhibition of C6 cells accurred mainly at G1 and S phase which were dose-and-time dependent.Conclution:HSP70 antisense oligomer can induce apoptosis and inhibit the growth of C6 cells by abrogating its HSP70 expression,HSP70 may be used as a target for the gene therapy of neuropongioma carcinoma.
