重组突变型IκBα cDNA对CNE1鼻咽癌细胞凋亡的诱导作用

【摘要】 目的:探讨突变型IκBα cDNA对CNE1鼻咽癌细胞凋亡的作用。方法:应用逆转录-聚合酶链式反应技术(RT-PCR)首先从CNE2鼻咽癌细胞中扩增突变型IκBα cDNA并将其克隆到pCLXSN逆转录病毒载体。然后,用脂质体介导的方法将含有该突变型IκBα cDNA的重组逆转录病毒载体转移到CNE1鼻咽癌细胞并用AnnexinⅤ-FITC细胞凋亡检测试剂盒和流式细胞术分析转染细胞凋亡情况。结果:通过RT-PCR技术,从CNE2鼻咽癌细胞扩增到IκBα cDNA并成功将其克隆到pCLXSN载体。将该重组逆转录病毒载体转染CNE1细胞约48小时后,AnnexinⅤ阳性细胞数为66.67%,而仅转染空白pCLXSN载体的CNE1细胞,An-nexinⅤ阳性细胞数为4.1%。结论:突变型IκBα cDNA能够诱导CNE1鼻咽癌细胞凋亡的发生。

【Abstract】 Objective: To explore the induction of apoptosis in CNE1 cells of nasopharyngeal carcinoma (NPC) by recombinant mutant IκBα cDNA. Methods: The mutant IκBα cDNA from CNE2 cells of NPC was firstly amplified by reverse transcription -polymerase chain reaction (RT-PCR) followed by cloning it into the retroviral vector of pCLXSN . Subsequently, the recombinant retroviral vector carrying the mutant IκBα cDNA was tansfected into CNE1 cells of NPC mediated by lipofectamine and apoptosis was then analyzed by flow cytometry with annexinⅤ-FITC apoptosis detection kit. Results: By RT-PCR, the mutant IκBα cDNA from CNE2 cells of NPC was successfully amplified and cloned into pCLXSN vector. About 48 hours after transfection of the recombinant retroviral vector into CNE1 cells, annexinⅤpositive cells was 66.67%. In contrast, the annexinⅤpositive cells was 4.1% in CNE1 cells of tansfected only with blank vector of pCLXSN. Conclusion: Apoptosis of CNE1 cells of NPC could be induced by the mutant IκBα cDNA.
