胰岛素样生长因子-1促进移植颗粒脂肪组织血管增生的实验研究

【摘要】 目的用成年雄性新西兰大白兔30只研究胰岛素样生长因子-1(insulin-like growth factors 1,IGF-1)在颗粒脂肪移植中的促进血管增生作用。方法取成年雄性新西兰大白兔腹部脂肪制备成颗粒脂肪,实验组经20 pg/L IGF-1处理和对照组经纯水处理后移植于背部。移植组织在不同观察时间取出后行α肌动蛋白(Actin)染色,用计算机图像分析技术测定微血管数目。结果IGF-1处理组血管数目在移植10、20、40 d后分别为93.41/cm2、138.12/cm2、104.15/cm2,显著高于同一观察时间的纯水处理组(P<0.05)。Actin染色强度也较高。结论一定浓度的IGF-1对移植颗粒脂肪组织具有促进血管增生作用。

【Abstract】 Objective To investigate the angiogenic effect of insulin-like growth factor 1(IGF-1) on grainy fat transplantation.Methods Grainy fat was obtained from the rabbit subcutaneous tissue in the abdomen.It was then mixed with 20 pg/L IGF-1 and distilled water and transplanted in the back subcutaneously.The transplanted tissue was taken out after 10,20 and 40 days and was stained with actin immunohistochemistry.The microvessel quantity was measured by medical imaging analysis system.Results The values of microvessel quantity in IGF-1 groups were 93.41/cm2 after 10 days,138.12/cm2 after 20 days and 104.15/cm2 after 40 days.This was obviously high compared with distilled water group in the same observing periods(P<0.05).Actin staining in IGF-1 group was also intense.Conclusion IGF-1 may be a candidate angiogenesis factor in grainy fat transplantation.
