Advances in Inhibition of Replication and Gene Expression of Hepatitis B Virus by RNA Interference
【Abstract】 RNA interference(RNAi) is a newly discovered phenomenon of post-transcriptional gene silence which is provoked by dsRNA. The dsRNA is initially cleaved by Dicer into 21-23 nt small interfering RNA(siRNA) and can then specifically target homologous mRNA for degradation by cellular ribonucleases. RNA interference is a common mechanism of gene regulation in biosystem evolutionary process. Current therapies for chronic hepatitis caused by hepatitis B virus (HBV) are limited. RNA interference may constitute a new therapeutic strategy for various hepatitis virus infections. In this review the advances in inhibition of HBV by RNAi, the unresolved questions and the therapeutic potential of RNAi were presented.
