两种茶树β-葡萄糖苷酶基因cDNA在原核中表达的研究

【摘要】 采用原核表达系统pET-32a(+)载体和BL21trxB(DE3)菌株对两条茶树β-葡萄糖苷酶基因cDNA(GluⅠ和GluⅡ)进行了表达,研究了不同诱导时间、不同诱导温度对两个茶树β-葡萄糖苷酶表达量的影响并测定了两者的活性。表达分析结果表明,诱导6h、25℃时,GluⅠ在总蛋白中所占比例最高;诱导6h、37℃时,GluⅡ在总蛋白中所占比例最高,活性测定结果表明,两种表达蛋白均具有正常的β-葡萄糖苷酶活性,在相同的测定条件下,GluⅡ活性(0.310±0.03U·ml-1)比GluⅠ(0.234±0.03U·ml-1)活性更高。

【Abstract】 Prokaryotic experimental system of expression vector pET-32a(+)and BL21trxB(DE3)strain were employed to express two kinds of tea beta-glucosidases(GluⅠand GluⅡ)cDNA gene.The impacts of different induction time and temperature on the expression amount of these two tea beta-glucosidase were studied and the activities of both beta-glucosidases were determined.The expression results revealed that the Glu I shown the highest proportion of the total protein in the conditions of six hours at 25℃,and then Glu II shown the highest proportion at six hours at 37℃.Both the resulted products exhibited normal beta-glucosidase activity,while Glu II(0.310±0.03U·ml-1)expressing product shown much higher beta-glucosidase activity than that of Glu I(0.234±0.03U·ml-1).
