猪SOCS-3 cDNA序列克隆及在各组织中表达
Cloning and Expression of Porcine SOCS-3 cDNA in Different Tissues
【摘要】 根据大鼠细胞因子信号转导抑制因子(suppressorofcytokinesignaling,SOCS)-3设计并合成一对引物,从八眉猪皮下脂肪组织提取总mRNA,RT-PCR扩增获得猪SOCS-3cDNA;利用半定量(semi-quantitative,SQ)RT-PCR技术检测大白猪各组织中SOCS-3mRNA的表达量.扩增获得SOCS-3基因GenBank登陆号DQ644577,用CDD软件进行SOCS-3基因结构分析,该序列具有SOCS-3特有的保守结构域SH2和SOCS-box.用ClustalW软件进行同源性分析发现,猪SOCS-3基因与人、大鼠和小鼠的同源性分别为92%、88%和88%.SQRT-PCR组织表达检测表明SOCS-3基因在心脏、肝脏、脾脏、肺脏、肾脏、肌肉、皮下脂肪和内脏脂肪中均有表达,其中肺脏和脾脏的表达丰度最高,肝脏和内脏脂肪中的表达量最低.
【Abstract】 A pair of PCR primers were designed and synthesized according to rat’s SOCS-3 gene sequence. Total RNA was isolated from adipocytes of Bamei pig and reverse-transcripted to cDNA, which was then subjected to SQ(semi-quantitative) RT-PCR. RT-PCR product was sequenced and submitted to GenBank(access number DQ644577). The SOCS-3 cDNA sequence was analyzed by Conserved Domain Database(CDD) and ClustalW software respectively. The results showed that the sequence contained two conservative regions(SH2 and SOCC-box) and had 92% homology to that of human,88% homology to that of rat and 88% to that of mouse. Also, we found that SOCS-3 gene was widely expressed in the heart,liver, spleen,lung,kidney,muscle,subcutaneous adipose and visceral adipose tissues. Its expression level was relatively higher in lung and spleen and lower in live and visceral adipose.
