PEG/(NH4)2SO4双水相萃取法提取果胶酶的研究
EXTRACTION OF PECTINASE USING AQUEOUS TWO-PHASE PARTITIONING
【摘要】 对PEG(聚乙二醇)(/NH4)2SO4双水相提取果胶酶进行了研究,考察了PEG分子量、pH、成相物质浓度、NaCl浓度、温度等因素对果胶酶分配系数的影响,结果表明:双水相最佳提取条件为PEG 1000 27%,(NH4)2SO419%,Na-Cl0.002%,pH5.0,温度50℃,在此条件下果胶酶分配系数可达到14.0。
【Abstract】 The partitioning of pectinase in polyethylene glycol (PEG)-(NH4)2SO4 aqueous two-phase systems was investigated, The influences of the molecular weight , pH, temperature and concentration of PEG, (NH4)2SO4 and NaCl on the partition coefficient k were studied. The optimal conditions of extraction showed as follow: the mass composition in two-phase aqueous system was PEG 1000 27 %,(NH4)2SO4 19 %,NaCl 0.002 %,pH 5.0,the highest partition coefficient k in above conditions could reach to 14.0.
