海参体壁粗蛋白酶的提取及酶学性质研究
STUDIES ON THE EXTRACTION AND ENZYMOLOGY CHARACTER OF THE CRUDE PROTEASE FROM THE BODY WALL OF HOLOTHURIAN
【摘要】 研究了海参体壁粗蛋白酶的提取方法和酶学性质,最佳提取条件为紫外线下照射30min,硫酸铵饱和度梯度为40% ̄60%。粗蛋白酶的最适pH值为5.0,最适反应温度为50℃,酶在50℃以下稳定性良好。在pH1.0 ̄7.0范围内,Ca2+对酶有激活作用,Mn2+、Cu2+对酶有抑制作用。提取的海参体壁蛋白酶是一种含有巯基的,类似胰蛋白酶的酶类。
【Abstract】 This article studied on the extraction and enzymology character of the crude protease from the body of Holothurian. The optimum extract condition was obtained as follows: the time of ultraviolet radiation was 30 min, the gradiet of ammonium sulfate was 40 %~60 %. The optimum pH was 5.0, the optimum temperature was 50 ℃, the crude protease was stable under 50 ℃. In the range of pH 1.0~7.0, Ca2+ activated the crude protease and Mn2+,Cu2+ inhibited the activity of the crude protease. So the crude protease from the body wall of Holothurian was considered a kind of protease containing thiol and like trypsin.
