聚酰胺分离纯化葡萄籽原花青素及部分产物组分构成的初步研究

【摘要】 以聚酰胺柱层析方法研究了上样流速、洗脱流速、丙酮浓度对聚酰胺吸附和洗脱原花青素的影响,并以HPLC-MS联检技术初步研究了20%和40%丙酮洗脱片段中的成分构成。结果显示,上样流速为0.44mL/min,在洗脱流速约为1mL/min条件下以80%丙酮洗脱时聚酰胺对原花青素的吸附和洗脱效果较好;以HPLC-MS技术在20%丙酮洗脱片段中检出儿茶素、表儿茶素和两种二聚体,而在40%丙酮洗脱片段中检出四种二聚体。

【Abstract】 The effect of sample loading,eluting velocity,as well as acetone concentration of on adsorption and desorption of proanthocyanidins in Polyamide column chromatography were studied.Parts of the composition in separated fractions were determined by HPLC-MS.The suitable conditions were loading up with 0.44 mL/min crude grape seed extract and eluting with 80 % acetone at the eluting velocity of 1 mL/min.Catechin,epicatechin,and tow dimmers were determined in the fraction of 20 % acetone by PR-HPLC.While four dimmers were determined in the fraction of 40 % acetone.
