酿酒酵母细胞破碎释放乙醛脱氢酶的研究

【摘要】 乙醛脱氢酶(ALDH)是一种胞内酶,在从酵母中分离提取ALDH的过程中,破碎细胞是其中的关键步骤之一。本文着重考察了搅拌粗提、珠磨破碎、高压细胞破碎仪和高压破碎仪的细胞破碎方法对释放ALDH的影响。结果表明,用高压破碎仪破碎,酵母细胞破碎率可达到95%以上,而且有效地防止了ALDH的分解及降解,是一种较为理想的破碎方法。

【Abstract】 Aldehyde dehydrogenase is a kind of endoenzyme,and cell disruption is the major step in its extraction from Saccharomyces ceravisia.The stirring,the glass bead breaker,the high pressure cell cracker and the high pressure cracker to break cell for releasing ALDH were investigated and compared.The results showed that cell disruption via high press cracker was the best method.The cell fragmentation efficiency of more than 95% could be reached via the high pressure cracker,and decomposition and degradation of ALDH were also prevented.
