沙棘果中黄酮糖苷和黄酮配基的测定

【摘要】 本文对沙棘果用甲醇进行了提取和盐酸催化的水解实验,采用HPLC方法测定了提取液和水解液中的黄酮配基含量。实验证明:用甲醇在室温下提取26h,每克果粉得到槲皮素0.025mg,山奈素0.015mg,异鼠李素0.035mg;而用盐酸对果粉水解3h;每克果粉能得到槲皮素0.383mg,山奈素0.025mg,异鼠李素0.992mg。提取的黄酮配基的数量分别是水解的6.52%,60.2%和3.52%。由此可见,沙棘果中的黄酮配基主要是以黄酮糖苷的形式存在,在水解过程中可由糖苷经水解反应形成,对槲皮素和异鼠李素尤其如此。

【Abstract】 The extraction and acid-catalyzed hydrolysis of sea buckthorn berries were reported in this work.The contents of flavonol aglucones in solutions obtained from the extraction and the hydrolysis were determined by HPLC.It was experimentally found that the contents of quercetin,kaempferol,and isorhamnetin in one gram of sea buckthorn berry were 0.025,0.015 and 0.035 mg,respectively.At the same time,during three hours the acid-catalyzed hydrolysis of one gram fruit gave 0.383mg of quercetin,0.025mg of kaempferol,and 0.992mg of isorhamnetin.The ratio of aglucone obtained by the extraction to that by the acid-catalyzed hydrolysis was 6.52%,60.2% and 3.52%,respectively.It is concluded that the main part of aglucone in sea buckthorn berry is in the form of flavonol glucosides and the free aglucone can be obtained only after the acid-catalyzed hydrolysis,especially for quercetin and isorhamnetin.
