人型支原体的分离培养和生长特性的研究
Study on the isolation and growth character of Mycoplasma hominis
【摘要】 目的研究人型支原体(Mh)的生长曲线、菌落形态演变等生物学特性及临床分离方法。方法取临床妇科阴道标本58例,同时接种液体培养基与固体培养基。在固体培养基上挑单菌落放入液体培养基进行纯化。选一株纯化的Mh接种于液体培养基,每隔4 h看颜色变化测pH值,并取10μL液体滴种在固体培养基上,计数菌落得出生长曲线。对不同生长时间的菌落拍照。从生长曲线得知对数生长期,并可在此期间进行保种。结果妇科阴道标本临床分离的Mh阳性率为8.62%;明确了在固体培养基上Mh菌落的生长演变过程;得到了Mh特有的生长曲线,并由此推出Mh在液体中最大生长数量及其时间、繁殖一代所需的时间和对数生长期及液体生化反应的pH值颜色改变规律。结论Mh的分离诊断,应该以固体培养基为基础,显微镜下看到典型的Mh特征性“油煎蛋”菌落为准。在液体培养基中Mh的传代以及保种在40 h左右,即1/2对数生长期最好。
【Abstract】 Objective To study the growth character and clinical isolation method of Mycoplasma hominis(Mh). Methods 58 clinical specimens were inoculated in liquid and solid culture media to calculate positive rate.The single Mh colony was picked into liquid medium to purify.Mh selected was inoculated into liquid medium.Every 4 hours we observed color and measured pH of the liquid medium and took 10 μL to drop it on solid medium.The colony was counted to draw the auxodrome.The colony morphous of different period was observed and photographed.The exponential phase of growth was obtained from the curve and the time of conserving the strain was defined. Results The positive rate of clinical isolation was 8.62%.The growth curve was procured from the number of colony.And further the Mh maximum concentration in the liquid,the time of reproducing one generation and the change regulation of liquid medium color were obtained.Successive colony evolution course on solid medium and the pH value of liquid medium were measured. Conclusions The use of liquid color change is unsuitable for Mh as judging the positive speciment.It colony should be based on solid medium,and on the classical morphous like "fried egg" under microscopic observation.
