TGF-β及其伙伴分子Smad4在大肠癌组织中的表达
Correlation of TGF-β and Smad4 in colonrectal carcinoma
【摘要】 目的探讨转化生长因子-β(TGF-β)及其伙伴分子Smad4基因在大肠癌组织中的表达。方法利用RT-PCR技术测定大肠癌组织TGF-β mRNA和Smad4 mRNA水平,并分析二者与大肠癌患者临床病理的关系。结果Smad4 mRNA在大肠癌组织中的阳性表达率明显下降。Smad4与大肠癌组织的分化程度、肿瘤大小、浸润深度、淋巴转移和TNM分期有关。TGF-β与大肠癌组织的浸润深度、淋巴转移和TNM分期有关。Smad4 mRNA在大肠癌组织分化程度高、肿瘤<5cm、浸润浅、无淋巴转移、TNM分期较早患者的阳性表达率高。TGF-β mRNA在大肠癌组织中的阳性表达率明显升高。TGF-β mRNA在浸润浅、无淋巴转移、TNM分期较早的大肠癌患者中阳性表达率低。大肠癌组织中TGF-β与Smad4表达呈负相关。结论TGF-β和Smad4共同作用于大肠癌的发生、发展的各生物学行为。
【Abstract】 [Objective] To explore the correlation of Smad4 mRNA and TGF-β mRNA in colonrectal carcinoma.[Methods] The levels of Smad4 mRNA and TGF-β mRNA were measured by reverse transcriptase polymerase chain reaction (RT-PCR) from 69 colonrectal carcinomas and 37 normal controls. The correlation of Smad4 mRNA and TGF-β mRNA was statisticaly analysis.[Results] The positive percentage of Smad4 mRNA in colonrectal carcinoma were lower than those of normal controls. The positive percentage of TGF-β mRNA in colonrectal carcinoma were higer than those of normal controls.The expression of Smad4 mRNA and TGF-β mRNA were all related to infiltration,lympy node metastasis and TNM stage. Negative correlation occurred in Smad4 mRNA and TGF-β mRNA.[Conclutions] TGF-β and Smad4 may be associated with the carcinogenesis, biological behaviors and prognosis of colonrectal carcinoma.
