新生期大鼠反复痫性发作模型的建立

【摘要】 目的:建立用于研究新生期大鼠反复痫性发作对其形态学及行为学影响的动物模型。方法:采用20只生后1 d的Wistar大鼠,在生后1、4、7 d给予腹腔注射匹罗卡品,另取12只生后1 d的Wistar大鼠作为对照组,在相应时间点腹腔注射生理盐水,采用5-溴脱氧尿苷(BrdU)标记新生细胞,观察新生鼠的行为学表现和组织学改变。结果:惊厥组新生鼠随着发作次数的增加,发作潜伏期变短,所需匹罗卡品的剂量变小,发作时间延长,发作强度变强,惊厥组3次发作后即刻神经发生较相应对照组明显减少,发作后45 d,惊厥组较对照组神经发生增加,但均未见明显的神经元丢失。结论:本实验建立的新生期大鼠反复痫性发作模型,为研究新生期反复惊厥发作对新生儿近期及远期的影响提供了一种可靠的动物模型。

【Abstract】 Objective:To establish a rat model of recurrent neonatal seizures,and evaluate its morphological and behavioral effects.Methods: The rats in this study were divided into an experimental group(n=20) and a control group(n=12).Neonatal Wistar rats(n=20) of the experimental group were subjected three times to pilocarpine injections intraperitoneally at postnatal day 1,4 and 7.Neonatal rats(n=12) of the control group were given a saline injection intraperitoneally at the same time.All rats were administered bromodeoxyuridine(BrdU) 36 hours before killing to label new-born cells.The rats′ behavioral and histological changes were observed at different times.Results: With an increase of seizure number,the mean time to onset and the dosage of pilocarpine decreased,and the manifestations of seizures became more intense.No seizure and death were found in the control group. BrdU-labeled cells decreased significantly in the experimental group compared with the controls at P7(P<0.05),while at P52 BrdU-labeled cells increased significantly in the experimental group(P<0.05).There was no evident neuron loss in either the experimental or the control groups at P7 and P52.Conclusion: The established recurrent seizures model neonatal rat is reliable for investigating the temporary and chronic effects of neonatal seizures.
