真菌提取物JN219抗病毒谱的初步研究
Antivirus spectra of JN219 extracted from certain strains of fungi
【摘要】 目的:探讨真菌提取物JN219的体外抗病毒谱。方法:采用体外细胞病变法,研究JN219对疱疹病毒科病毒(HCMV、HSV-1、HSV-2)、腺病毒科病毒(Ad3)、副粘病毒科病毒(RSV)、呼肠病毒科病毒(RVSA11)、正粘病毒科病毒(流感甲地方株、流感乙地方株)、小RNA病毒科病毒(COX B1-6)、弹状病毒科病毒(VSV)共15株病毒的抗病毒活性。将JN219与以上病毒作用1 h后接种于相应宿主细胞(同时设有病毒对照和细胞对照),当病毒对照病变达到90%以上时终止培养,宿主细胞用1%中性红染色,在450 nm处读取吸光度A值,计算细胞存活率、半数中毒浓度(TD50)、半数有效浓度(IC50)和抑毒指数(TI)。并根据TI判断JN219的抗病毒活性,确定其抗病毒谱。结果:JN219对HCMV、RV(SA11)、VSV有抑制作用,其IC50分别为1.42、0.96和1.22μg/ml,TI分别为35.26、52.30、40.82;对HSV-1、HSV-2、RSV、流感病毒甲、流感病毒乙有部分抑制作用;对COX B1-6、腺病毒(Ad3)无作用。结论:JN219抗病毒谱比较广泛。
【Abstract】 Objective: To investigate the antivirus spectra of singlet compound named JN219 extracted from certain strains of fungi.Methods: Fifteen different strains of viruses,including herpesviridae virus(HCMV,HSV-1,HSV-2),adenoviridae virus(Ad3),paramyxoviridae virus(RSV),reoviridae virus(RV SA11),orthomyxoviridae(influenza A,influenza B),picornaviridae(COX B1-6)and rhabdoviridae(VSV)were used for in-vitro antivirus experiments,and the antiviral activity and the cytotoxicity of JN219 were evaluated by the cytopathic effect(CPE) method.Infected cells were dyed with 1% neutral red,and the decolored and the absorptance A were read by an enzyme label meter.The cell survival rate,the median toxic concentration,the median inhibiting concentrations and therapeutic indexes were calculated by the Reed-Muench method.Antivirus stability experiments of JN219 were carried out at the same time.Results: Cytopathic effects caused by HCMV,VSV and RV SA11 were significantly inhibited.The median inhibiting concentrations(IC50) were 1.42,0.96 and 1.22?μg/ml,respectively,and the therapeutic indexes(TI) were(35.26,) 52.30 and 40.82,respectively.HSV1,HSV-2,RSV,influenza A and influenza B were partially inhibited,however COX B1-6 and Ad3 were not inhibited.The cytotoxicity of the samples appeared only in the first dilution and the antiviral activity was unstable.Conclusion: JN219 has a wide antivirus spectra.
