肾阳虚证患者血液生化、免疫指标与基因表达谱结果的对比研究

【摘要】 目的:对比研究肾阳虚寒证患者血液生化、免疫指标以及差异基因表达谱的临床应用价值。方法:针对一个典型肾阳虚家系15个成员,运用现代检测技术进行血液生化、免疫学指标检测,同时采用人类Oligo芯片杂交技术对其外周血单个核细胞的差异基因表达谱进行分析研究。结果:虽然每个成员分别出现了2-8项异常血液学指标,但整体而言,该家系肾阳虚患者与正常人之间在糖、蛋白质、脂类代谢上和体液、细胞免疫方面无明显差异(P>0.l);而5张芯片的杂交结果显示,共有53个基因反复出现在差异基因表达谱之中,这些基因可能在肾阳虚寒证的发生发展中起着关键性作用。结论:常规血液生化、免疫学指标对肾阳虚寒证的参考意义不太大,而芯片检测技术以及由此获得的差异表达基因谱在中医证候研究中具有潜在的临床应用价值。

【Abstract】 Objective:To study comparatively the potential clinical application of biochemical index,immunological index and differential expression gene profile on Kidney Yang-deficiency syndrome,a complicated Chinese medical syndrome.Methods:A total of 15 patients were chosen from a typical family with Kidney Yang-deficiency syndrome.Relevant assays were employed for blood serum tests.In order to indicate the differential expression gene profiles,researchers utilized the Human Oligo chips to conduct microarray analysis.Results:Although there were two to eight biochemical or immunological indices abnormal in each patient(P>0.1),no statistical differences were found in the metabolism of glucose,protein and lipid,and humoral immunity and cellular immunity in patients with Kidney Yang-deficiency syndrome compared with the standard references.According to the results of 53 microarray test chips,there were 5 genes displayed repeatedly in the differential gene expression profiles,which might play a critical role in the development and progress of Kidney Yang-deficiency syndrome.Conclusion:The researches imply that the routine tests of either biochemistry or immunology are of little value for the diagnosis of Kidney Yang-deficiency syndrome.However,microarray analysis and the subsequent differential gene expression profiles may have some potential values on the study of Kidney Yang-deficiency syndromes.
