两种杂合抗菌肽在毕赤酵母中的表达及活性测定
Secreted expression and activity detection of two hybrid antimicrobial peptides in Pichia pastoris
【摘要】 参照毕赤巴斯德酵母(Pichia pastoris)的偏好密码子,改造克隆杂合抗菌肽CA(1~7)M(4~11)和CB(1~7)M(4~11)基因,将改造后的基因克隆到pPICZ-αA载体中,构建分泌型表达载体pPICZ-αA-CAM和pPICZ-αA-CBM,转化宿主菌Pichia pastorisX-33,在甲醇的诱导下进行表达.实验结果表明,杂合抗菌肽获得表达,两种表达产物具有明显的抗菌活性.此外,本文还比较了二者的抗菌谱,分析了杂合肽的性质,优化了酵母电转化条件.
【Abstract】 According to the partiality codon of Pichia pastoris,hybrid antimicrobial peptide CA(1~7)M(4~11) and CB(1~7)M(4~11) gene were reconstructed,synthesized and cloned into pPICZα-A,the recombinant expression vectors were transformed into host strain Pichia pastoris X-33.Based on the experimental result,hybrid antimicrobial peptides were expressed,which showed obvious antibacterial activity.Furthermore,the antibacterial spectrums and properties of the hybrid antimicrobial peptides were analyzed,the conditions of yeast electrotransformation were optimized.
